A CONGO RED STAINING METHOD FOR EPOXY-EMBEDDED AMYLOID

A Congo red staining method for osmium-fixed, epoxy-embedded tissues is described. The staining is carried out by floating thick sections (0.5-2.0 µ) upon a drop of 0.5-1.0% Congo red in 0.1 N sodium hydroxide at 45°C for 60-120 min. The amyloid, which stains a clear red, shows characteristic green birefringence when viewed in the polarization microscope. This staining method is useful for identification and mapping of amyloid substance in serial thick sections and for sampling in preparation for the electron microscopic studies of adjacent thin sections.
