Diffusion of extracellular free rhodamine 110 into cells. The substrate Arg-Nle-Nle-2-R110 was incubated with control lymphocytes for 8h at 8C. The lymphocytes were centrifuged and the cell supernatant (SN Arg-Nle-Nle-2-R110) was collected and incubated with fresh lymphocytes for additional 8h. TPP1 -/- lymphocytes (A) and control lymphocytes (B) merely show a slight increase in fluorescent signal intensity (dotted line) when compared to lymphocytes incubated with buffer solution only (solid line).
