Expression of ABCA3 in Developing Lung and Other Tissues

  • Supplemental Figure 1 - Specificity of rabbit polyclonal antibody generated to ABCA3. A panel of adult and fetal mouse tissues was probed with preimmune, immune, and antigen-absorbed immune rabbit serum, as described in Materials and Methods. Little-to-no background staining was detected with the preimmune serum (A, D, G, H). Likewise, no specific immunostaining was detected in adult or fetal tissues with the preimmune serum (A, D, G, H). Specific staining for ABCA3, visualized as a black cellular precipitate, was detected only in adult lungs (B), fetal lungs at E18.5 (H) and E 17.5 (K), and in Purkinje cells of the cerebellum (E). Preincubation of the antibody with increasing concentration (5-200 µg) of the ABCA3 peptide, blocked specific staining in the lung (C, I, J) and in the cerebellum (F). Scale bar equals 100 µm for all panels.
  • Supplemental Figure 2 - Specificity of the rabbit polyclonal antibody generated to ABCA3 for the detection of the protein in human tissues. Preincubation of the ABCA3 antibody with its antigenic peptide, blocked specific staining in human lung (A, B), renal tubules (C, D) and pancreas (E, F). Immunostaining for ABCA3 is seen on the left (A, C, E) and after preincubation with the peptide on the right (B, D, F).
