Western Analysis of total protein from human cells with J2. Total cell protein was extracted from NT2 and 293T cells, which do not express detectable levels of NAIP, were transfected with plasmid expressing full length NAIP. A; adsorbed antibody. A broad range molecular weight marker was used to determine sizes. Anticipated MW for NAIP: 158 kD. The lanes are: brain (1), heart (2), kidney (3), liver (4), lung (5), ovary (6), skeletal muscle (7), testis (8), spleen (9) and placenta (10).

NAIP expression in mouse lung assessed by immunohistochemistry. The tissue was obtained post-mortem and paraffin embedded. Sections were treated with antigen retrival system (ARS) prior to incubation with J2 (1/100), followed by a biotinylated secondary antibody and consecutive incubation with streptavidin-horseradish peroxidase complex and 3'3-diaminobenzindine (DAB), which results in a brown stain. Tissues were counterstained with hematoxylin (blue stain). NAIP immunoreactivity in the trachea (A). NAIP expression in the adipocytes (B); expression of NAIP in the alveolar septae in the upper lung (C) and in the lower lung (D).
